| Abstract: | Phytochemical studies of the flowers of Thong-Kwaw (Butea monosperma) led to the isolation and identification of seven flavonoid compounds and their derivatives, namely medicarpin (1), sulfuretin (2), liquiritigenin (3), kaempferol (4), butein (5), iso-butrin (6), and butrin (7), using NMR spectroscopic techniques for structural analysis. The isolated compounds exhibited diverse biological activities. In melanin biosynthesis pathway, liquiritigenin (3) showed excellent tyrosinase inhibitory activity (IC50 = 13.07 ± 2.58 µM) via a competitive inhibition mechanism and reduced melanin production in MNT-1 cells by 41% at a concentration of 300 µM without cytotoxic effects. Additionally, butein (5) and sulfuretin (2) demonstrated strong antioxidant activities using DPPH, ABTS, and FRAP assays and butein (5) exhibited anti-inflammatory activity through potent inhibition of nitric oxide (NO) production. In antibacterial and anticancer activity tests, medicarpin (1) showed the most potent activity against Cutibacterium acnes (MIC = 30 µM) and significantly inhibited biofilm formation by both C. acnes and Staphylococcus aureus. For anti-cancer activity, sulfuretin (2) and butein (5) displayed moderate effects against HCT116 colon cancer cells, while kaempferol (4), butein (5), and iso-butrin (6) were active against HeLa cervical cancer cells. In conclusion, this study highlights the significant potential of B. monosperma flower extracts, providing valuable insights for future applications in the cosmetic and pharmaceutical industries. Potential of B. monosperma flower extracts, providing valuable insights for future applications in the cosmetic and pharmaceutical industries. การศึกษาองค์ประกอบทางพฤกษเคมีจากดอกทองกวาว (Butea monosperma) นำไปสู่การแยกและพิสูจน์เอกลักษณ์ของสารประกอบฟลาโวนอยด์และอนุพันธ์รวม 7 ชนิด ได้แก่ medicarpin (1), sulfuretin (2), liquiritigenin (3), kaempferol (4), butein (5), iso-butrin (6) และ butrin (7) โดยใช้เทคนิคทางสเปกโทรสโกปี NMR ในการวิเคราะห์โครงสร้าง สารประกอบที่แยกได้แสดงฤทธิ์ทางชีวภาพที่หลากหลาย จากผลการทดลองในระดับเซลล์ที่เกี่ยวข้องกับกระบวนการสร้างเม็ดสี พบว่า liquiritigenin (3) แสดงฤทธิ์ยับยั้งเอนไซม์ไทโรซิเนสได้อย่างดีเยี่ยม (IC50 = 13.07 ± 2.58 µM) โดยมีกลไกการยับยั้งแบบแข่งขัน (competitive inhibitor) และสามารถลดการสร้างเม็ดสีในเซลล์ MNT-1 ได้ถึง 41% ที่ความเข้มข้น 300 µM โดยไม่แสดงความเป็นพิษต่อเซลล์ ในขณะที่ butein (5) และ sulfuretin (2) แสดงฤทธิ์ต้านอนุมูลอิสระที่สูงสุดในการทดสอบด้วยวิธี DPPH, ABTS และ FRAP นอกจากนี้ butein (5) ยังแสดงฤทธิ์ต้านการอักเสบผ่านการยับยั้งไนตริกออกไซด์ (NO) ได้อย่างมีประสิทธิภาพสูงสุด ในการทดสอบฤทธิ์ต่อเชื้อแบคทีเรียก่อโรคและเซลล์มะเร็ง medicarpin (1) แสดงฤทธิ์ต้านเชื้อแบคทีเรีย Cutibacterium acnes ได้ดีที่สุด (MIC = 30 µM) และที่สำคัญคือสามารถยับยั้งการสร้างไบโอฟิล์มของทั้งเชื้อ C. acnes และ Staphylococcus aureus ได้อย่างมีนัยสำคัญ สำหรับฤทธิ์ต้านเซลล์มะเร็ง พบว่า sulfuretin (2) และ butein (5) มีฤทธิ์ระดับปานกลางต่อเซลล์มะเร็งลำไส้ใหญ่ HCT116 ในขณะที่ kaempferol (4), butein (5) และ iso-butrin (6) มีฤทธิ์ต่อเซลล์มะเร็งปากมดลูก HeLa งานวิจัยนี้ได้ชี้ให้เห็นถึงศักยภาพของสารสกัดจากดอกทองกวาว และข้อมูลพื้นฐานสำคัญในการต่อยอดสู่การประยุกต์ใช้ในอุตสาหกรรมเวชสำอางและยาต่อไป |
